 Background: Exercise training, resveratrol supplementation, and cellular aging are important issues in nonalcoholic fatty liver disease. The present study aimed to investigate the effect of 8 weeks of aerobic training and resveratrol consumption on the expression of some indicators of cellular aging in the liver tissue of a nonalcoholic fatty liver disease rat model.
Methods: In the present experimental study, 35 eight-week-old male Wistar rats were randomly divided into two groups: patients with nonalcoholic fatty liver disease and a control group. Then, the rats in the control group were placed on a standard diet for 6 weeks, while to induce nonalcoholic fatty liver disease in the rats, the animals were placed on a high-fat diet for 6 weeks. The rats in the patient group were further divided into four experimental subgroups: non-alcoholic fatty liver, non-alcoholic fatty liver-exercise, non-alcoholic fatty liver-resveratrol, and non-alcoholic fatty liver-exercise-resveratrol. The rats in the exercise group underwent an eight-week (5 days/week) aerobic interval-training program. Rats in the supplement group were injected intraperitoneally with 20 mg/kg of resveratrol per day for 8 weeks.
Results: The results showed a significant increase in the rate of changes in p21 expression in the non-alcoholic fatty liver (p = 0.0001), non-alcoholic fatty liver-exercise (p = 0.002), and non-alcoholic fatty liver-resveratrol (p = 0.002) groups compared to healthy controls, and also a significant increase in the rate of changes in p16 expression in the non-alcoholic fatty liver (p = 0.0001), non-alcoholic fatty liver-exercise (p = 0.006), and non-alcoholic fatty liver-resveratrol (p = 0.003) groups compared to healthy controls.
Conclusion: Treating nonalcoholic fatty liver disease with exercise and resveratrol improves markers of liver aging in rats. Eliminating or reducing cellular markers using exercise and resveratrol may be an effective strategy for improving liver damage associated with nonalcoholic fatty liver disease.
