fa ارتباط پلی‌مورفیسم rs2236242 ژن واسپین با بیماری کبد چرب غیرالکلی ASSOCIATION OF VASPIN RS2236242 GENE POLYMORPHISM WITH NON-ALCOHOLIC FATTY LIVER DISEASE بیوشیمی بیوشیمی پژوهشي(توصیفی- تحلیلی) Research <w:sdt docpart="19BD721496B24FB3B1EFAE808B42E267" id="1932386431" prefixmappings="xmlns:ns0='http://purl.org/dc/elements/1.1/' xmlns:ns1='http://schemas.openxmlformats.org/package/2006/metadata/core-properties' " storeitemid="X_6C3C8BC8-F283-45AE-878A-BAB7291924A1" text="t" title="Subject" xpath="/ns1:coreProperties[1]/ns0:subject[1]">پیش‌زمینه و هدف</w:sdt>: بیماری کبد چرب غیرالکلی (NAFLD) شایع‌ترین بیماری کبدی و یک مشکل مهم بهداشت عمومی در سراسر جهان است. شیوع NAFLD به‌طور پیوسته در حال افزایش است که با عوارض جدی مانند سیروز و کارسینوم کبدی همراه است، همچنین با سندرم مقاومت به انسولین و بیماری‌های قلبی عروقی مرتبط است. در این مطالعه، ارتباط پلی‌مورفیسم rs2236242 ژن واسپین با NAFLD بررسی شده است. مواد و روش کار: این مطالعه مورد-شاهدی (نمونه‌گیری تصادفی) بر روی 150 شرکت‌کننده، شامل 75 شرکت‌کننده با بیماری‌های کبد چرب غیرالکلی به‌عنوان مورد و 75 شرکت‌کننده به‌عنوان گروه عادی انجام شد. شرکت‌کنندگان با سابقه مصرف الکل، اختلالات متابولیک و سایر بیماری‌های کبدی، از مطالعه حذف شدند. واسپین و انسولین سرم با کیت الایزا و سایر متغیرها به روش استاندارد اندازه‌گیری شدند. تعیین ژنوتیپ‌های rs2236242 با استفاده از واکنش زنجیره‌ای پلیمراز تتراآرمز&zwj; انجام شد. تجزیه‌وتحلیل آماری با استفاده از نرم‌افزار SPSS نسخه 20 انجام شد. یافته‌ها: تفاوت معنی‌داری در سطح واسپین در بیماران مبتلا به NAFLD نسبت به افراد سالم یافت نشد (05/0≤P). سطح هیچ‌یک از متغیرها در افراد سالم و بیماران برای ژنوتیپ‌های TA و TT تفاوت معنی‌داری نداشت. بین پلی‌مورفیسم rs2236242 و مقاومت به انسولین ارتباط معنی‌داری وجود نداشت. هیچ ارتباطی بین این پلی‌مورفیسم و NAFLD یافت نشد (05/0≤P). بحث و نتیجه‌گیری: یافته‌ها نشان می‌دهد که بین پلی‌مورفیسم rs2236242 با سطوح واسپین، مقاومت به انسولین و بیماری کبد چرب غیرالکلی ارتباطی وجود ندارد. بااین‌حال، محدودیت‌هایی مانند حجم نمونه محدود و استفاده از سونوگرافی بر تعمیم‌پذیری یافته‌ها تأثیرگذار است.   <a name="_Toc416081061">Background & Aims:</a> Non-alcoholic fatty liver disease (NAFLD) is the most common liver disease and a major public health problem worldwide. NAFLD has been steadily increasing in prevalence and is associated with serious complications such as cirrhosis and hepatocellular carcinoma, It is also linked to insulin resistance syndrome and cardiovascular diseases. In this study, the association of vaspin rs2236242 gene polymorphism with NAFLD is investigated. Materials & Methods: The case-control study (random sampling) was conducted on 150 participants, includes 75 participants with non-alcoholic fatty liver diseases as cases and 75 participants as normal group. Participants with a history of alcohol consumption, metabolic disorders or other liver diseases were excluded from the study. The serum vaspin and insulin were measured by ELISA kit and other variables by standard method. Determining the genotypes of rs2236242 was done using the tetra arms polymerase chain reaction. Statistical analysis was performed using SPSS software version 20. Results: There was no significant difference in vaspin levels in the patients with NAFLD compared with healthy individuals (p ≥0.05). The levels of any of the variables in healthy individuals and patients for GG, AG, and AA genotypes were not significantly different. There was no a significant relationship between rs2236242polymorphism and insulin resistance. No association was found between this polymorphism and NAFLD (p ≥0.05). Conclusion: The findings showed that there is no relationship between rs2236242 polymorphism with vaspin levels, insulin resistance and non-alcoholic fatty liver disease.
