fa مطالعه هم‌ارزی زیستی برون تن سه برند ژنریک آسیکلوویر 400 میلی‌گرمی موجود در بازار دارویی ایران IN VITRO BIOEQUIVALENCE STUDY OF THREE GENERIC BRANDS OF ACYCLOVIR 400 MG TABLETS IN IRAN MARKET داروسازی داروسازی پژوهشي(توصیفی- تحلیلی) Research پیش‌زمینه و هدف: مطالعات هم‌ارزی زیستی برای ارزیابی تشابه درمانی دو فرمولاسیون مختلف از یک دارو (برند ژنریک و برند مرجع) طرح‌ریزی شده‌اند. هدف از مطالعه حاضر مطالعه‌ی هم‌ارزی زیستی برون تن بر روی سه برند ژنریک آسیکلوویر 400 میلی‌گرم موجود در بازار دارویی ایران است. مواد و روش‌ کار: آزمون‌های سنجش ماده مؤثره، تغییرات وزنی و انحلال بر روی سه برند ژنریک و برند مرجع Lovir)) انجام شد. نتایج این سه آزمون با استانداردهای دارویی فارماکوپه امریکا (USP) مقایسه شد. جهت مقایسه پروفایل انحلال و تعیین تشابه سه برند ژنریک با برند مرجع، فاکتورهای تفاوت (f1) و تشابه (f2) محاسبه شدند. یافته‌ها: نتایج تست سنجش ماده دارویی نشان داد که میزان ماده مؤثره در تمامی برندهای قرص آسیکلوویر در محدوده‌ی 90 الی 110درصد ادعای برچسب است. نتایج آزمون تغییرات وزنی نشان داد که تمامی برندها با مشخصات USP مطابقت دارند. نتایج تست انحلال نشان داد که همه برندها در بازه‌ی زمانی 45 دقیقه بیش از 80 درصد ماده‌ی مؤثره دارویی را آزاد می‌کنند که با مشخصات USP مطابقت دارند. در مورد پروفایل انحلال و محاسبه‌ی فاکتورهای تشابه (F1) و تفاوت (F2) هیچ‌کدام از برندهای ژنریک مشخصات رهش مشابهی را با برند مرجع نشان ندادند. بحث و نتیجه‌گیری: این مطالعه نشان داد که همه‌ی برندهای قرص اسیکلوویر دارای مشخصات کیفی موردقبول در آزمون‌های سنجش ماده، تغییرات وزنی و انحلال هستند ولی نمی‌توانند به‌عنوان جایگزین برند مرجع در نظر گرفته شوند.   <a name="_Toc416081061">Background &</a> Aims: Bioequivalence studies are established for evaluating the therapeutic equivalency of two different formulations (A generic formulation and a reference formulation) of the same drug. The aim of present study was to study in vitro bioequivalence of three brands of acyclovir 400 mg tablets in the Iran market. Materials & Methods: Assay, weight variation, and dissolution studies were performed for three generic brands and one reference brand (Lovir). These three tests were compared with the United States Pharmacopeia (USP) drug standards. In order to compare the dissolution profile and determine the similarity of three generic brands with the reference brand, the difference (f1) and similarity (f2) factors were calculated. Results: The active pharmaceutical ingredients quantitative assay showed that all the brands of acyclovir tablets were between 90% and 110% of label claim. The results of weight variation tests indicated that all samples complied with USP specification limits. The results of the dissolution test showed that all brands released more than 80% of active pharmaceutical ingredient in a 45 minutes period, which corresponded to USP specifications. Regarding the dissolution profile and calculation of similarity (f1) and difference (f2) factors, none of the generic brands showed the same drug release characteristics as the reference brand.
